A diet high in saturated fat and cholesterol contributes to obesity, which is a chronic factor that contributes to the onset of obesity and metabolic diseases. Hepatic oxidative stress is commonly reported in various animal models of obesity and is associated with increased NADPH oxidation. We have previously described several antioxidant mechanisms that use folic acid to protect against oxidative stress-induced hyperhomocysteinemia. The aim of this study was to find out whether folic acid supplementation improves hepatic oxidative stress induced by high fat and to determine the mechanism. Male C57BL/6J mice were fed a control diet, a high-fat diet, or a high-fat diet plus folic acid for 12 weeks. A high-fat diet induced weight gain, hepatic lipid peroxidation, and liver damage. There was a significant increase in liver NADPH oxidase activity, which was associated with increased expression of NADPH oxidase subunits. Folic acid supplementation has been shown to have a protective effect against high fat-induced hepatic oxidative stress and liver injury. The analysis revealed that the antioxidant effect of folic acid was mainly due to the regulation of NADPH oxidation. These results suggest that folic acid supplementation may be hepatoprotective against liver damage associated with a high-fat diet.

Hepatic NADPH oxidase activity was markedly increased, and was associated with increased expression of several subunits of hepatic NADPH oxidase.
